植物細胞の分化を担う液胞プロセシング系の分子機構

研究概要

高等植物の分化の特徴は細胞の柔軟性にあり,これを支えているのが可逆的なオルガネラの変換能力である.中でも液胞は植物の生長・分化の過程や環境の変化に応じて大きく変動するオルガネラである.液胞の分化は,植物の細胞分化や細胞死に重要な役割を果たしていることが明らかになってきた.本研究では,液胞タンパク質の機能発現に関わる液胞プロセシング系の解明を目的として研究を進めてきた.
(1)昨年度見いだした新規オルガネラERボディは幼植物体の表皮細胞に局在し,液胞プロセシング酵素などのストレス誘導型のプロテアーゼ前駆体を蓄積している.成熟葉にはERボディは全く検出されないが,傷害や虫害により誘導されることが分かった.ERbody形成不全の変異体の一つの原因遺伝子はbHLH型の転写因子であることが分かった.ERボディは植物の生体防御に関わるオルガネラであると考えられる.
(2)動物細胞のプログラム細胞死においてはCaspasesが重要な役割を果たしていることは良く知られており,植物で細胞にもこの活性が認められているにも関わらず,その実体は不明のままであった.タバコモザイクウィルス(TMV)に感染したタバコ葉は過敏感反応死を引き起こすが,このプロセスの極初期において液胞プロセシング酵素が機能していること,およびこのVPEがCaspase-1様活性を示すことが明らかとなった.
