機能性脂質の創製を目指した油脂生産微生物のメタボリックエンジニアリング

研究概要	高度不飽和脂肪酸生産菌Mortierella alpina 1S-4を代謝工学的に育種するために必須となる宿主ベクター系の構築を試みた。本年度は相同組み換え時のマーカー遺伝子となるorotidine-5'-phosphate decarboxylase遺伝子を取得するとともに、本菌のウラシル要求性変異株の取得に向け、本菌の胞子をニトロソグアニジン処理し、ピリミジン生合成系の中間物質(オロト酸)のアナログ5'-フルオロオロト酸を含む培地にて生育可能な変異株を取得した。また、M.alpina 1S-4の高度不飽和脂肪酸生合成遺伝子の取得を試み、Δ6不飽和化酵素アイソザイム遺伝子の取得に成功するとともに、Δ12不飽和化酵素活性低下株のΔ12不飽和化酵素遺伝子の取得に成功した。さらに.M.alpina 1S-4の鎖長延長酵素について知見を得るべく、鎖長延長酵素欠損変異株の取得を試み、変異株M1を得た。変異株M1では、16:0から18:0への鎖長延長能の低下により16:0が蓄積し、蓄積した16:0がΔ9不飽和化反応とそれに引き続く鎖長延長反応により16:1ω7を経由して18:1ω7へと変換される一方、一部の16:1ω7はn-7経路を経て18:3ω7(16:1ω7→16:2ω7→18:2ω7→18:3ω7)へ、あるいn-4経路を経て20:4ω(16:1ω7→16:2ω4→16:3ω4→18:3ω4→20:4ω4)へ変換された。一方、最近注目を集めている新規機能性脂肪酸・共役リノール酸をリノール酸の異性化を経て生産する菌株のスクリーニングを行った結果、高活性菌としてLactobacillus plantarum AKU1009a株を見いだした。また、アセチレン脂肪酸であるジヒドロクレペニン酸を生産する菌株として、Shizophillum commune YK5株を見いだした。
