ポリオウイルスの神経病原性と神経細胞機能の研究

研究概要

ポリオウイルス(PV)レセプター(PVR)のアイソタイプ(PVRα,PVRδ)間で、極性細胞内での局在性に違いがあることを発見した。PVRαの細胞質内ドメインのタイロシンモチーフには、clathrin adaptor complex AP-1Bのサブユニットμ1Bが結合することが確認された。このμ1Bはバソラテラルソーティングに関与する分子であり、PVRαはこの分子によって基底膜側へソーティングされると考えられた。
培養神経細胞においては、抗PV抗体または抗PVR抗体存在下、PV感染による細胞変性効果が起きないことを発見した。このとき、細胞側の蛋白合成はいったん減少するが回復し、反対にウイルス蛋白合成は、いったんは増加するがその後減少することを明らかにした。ウイルス蛋白質2AはCap依存的翻訳開始因子の一つeIF4Gの切断に関与することが知られているが、2Aの増減に相応してeIF4Gの切断型が増減していた。したがって、これまでのところ蛋白合成の段階で神経細胞の変性効果発現が制御されている可能性を考えている。
これまでにPVがPVR依存的にシナプスからエンドサイトーシスされ、運動神経軸索内を逆行性モーター蛋白質である細胞質ダイニンにより逆行性輸送されるという仮説を提示した。この仮説を証明するため、PVRの細胞質内ドメインとTCTEL1との結合を確認し、結合に関わるモチーフも同定した。このモチーフに変異を導入すると、PV存在下における小胞の逆行性軸索輸送系が不活発になることが小胞輸送の高速度撮影実験系により明らかになった。
BDNFを発現するPVベクターをPV感受性トランスジェニックマウスに接種したところ、BDNF分子は運動神経特異的に発現していた。このPVベクターは少なくとも3代目までは安定に存在していることが確認された。
