研究概要

アポトーシス過程におけるクロマチンDNA断片化の機構解明は現代生物学の大きな研究課題となっている。これに関与すると想定されるエンドヌクレアーゼとしてDNase IおよびDNase IIがあるが、これらがアポトーシスに果たす役割に関する実験的根拠はほとんど皆無である。本研究ではDNase IおよびDNase IIのアポトーシスへの関与機構を解明することを主な目的とした。
1.両生類DNase Iは他の脊椎動物酵素と大きく異なる性状を有する。両生類や爬虫類などの冷血動物から鳥類や哺乳類などの温血動物がどのように進化してきたかを明らかにするため、系統学上両生類と鳥類・哺乳類の中間に位置する爬虫類酵素は興味ある研究対象である。爬虫類DNase Iに関する解析結果から、哺乳類酵素の熱安定性は下記の2段階の変異を経て獲得されたと考えられた:(1)両生類から爬虫類への進化に伴う両生類酵素におけるSer205の欠失、(2)爬虫類から鳥類への進化に伴う爬虫類酵素におけるIle130のLeuへの置換。
2.胸痛発作直後に来院した急性心筋梗塞患者群と狭心症や不安定狭心症などの非心筋梗塞患者群について、その血中DNase I活性の経時的変動を精査した。狭心症および不安定狭心症患者群の血中DNase I活性は正常値内であり、経時的変動も認められなかった。心筋梗塞患者群においては、約9割の症例で胸痛発作直後に血中DNase I活性が上昇し、2ないし3時間後に最大値となり、その後活性は減少し、およそ12時間以内に正常値に復帰した。従って、血中DNase I活性は心筋発作直後に一過性の上昇を示すことが明らかとなった。他方、これまで心筋梗塞の診断に使用されてきたtroponin Tは発症直後には顕著な濃度の上昇を示さないことから、血中DNase I活性値上昇は急性心筋梗塞の超早期診断マーカーになりうると考えられた。
