研究概要	ベーチェット病、Vogt-小柳-原田病をはじめとした難治性炎症性眼免疫病は、その発症にHLAが深く関わっていることが示唆されている。したがって、HLA遺伝子領域の詳細な解析を行うため、また、ヒトゲノムプロジェクトの一環として、今年度、我々はHLAクラスI領域の全塩基配列、1.9Mb(1.9X106bp)を正確に決定した。既存のDNAデータバンクとのホモロジー解析およびGrail、Hexon解析の結果、23個の発現既知遺伝子、12個の発現新遺伝子、22個のESTと一致する領域、67個の偽遺伝子および3個の発現可能な塩基配列の計127個の遺伝子あるいは遺伝子候補領域を見出した。また、この膨大な塩基配列をもとに、この領域に存在するマイクロサテライト(反復配列)を検索した。その結果、2〜5塩基の反復配列からなる計758個のマイクロサテライトを見出した。これらの内70個について多型性を検索したところ、38個が高度な遺伝的多型性を有し、HLA領域に存在する多くの遺伝病の原因遺伝子の詳細なマッピングに有用な遺伝多型マーカーとなり得ると考えられた。したがって、日本人、ギリシャ人、イタリア人、ヨルダン人、イラン人ベーチェット病患者を対象として、これらのマイクロサテライト多型解析およびMICA遺伝子解析を行ったところ、HLA-B^51対立遺伝子が本病発症に第一義的に関与し、患者群で高度に保存されているMIC-A^009-HLA-B^*51ミニハプロタイプが本病発症に重要な役割を担っていることが示唆された。今後、全染色体を対象としたゲノムワイドなマイクロサテライト多型性解析が必要と考えられた。また、これらの本病関連候補遺伝子を導入したトランスジェニックマウス(Tg)の開発では、今年度、細胞表面でのHLA-B51蛋白の発現量を増加させるため、HLA-B51Tgとヒトβ2mTgの交配により、両遺伝子を持つTgを作製した。今後、このマウスとマウスβ2mノックアウトマウスとの交配を計画している。
