研究概要	研究代表者らはシェーグレン症候群の病因と密接に関連した責任抗原として分子量120KDの自己抗原α-フォドリンを分離精製した。そこで、自己抗原120KDα-フォドリンの成立機序を解明し、疾患特異的免疫応答の制御法・治療法の確立が急務とされてきた。本研究ではシェーグレン症候群の病因抗原120KDα-フォドリンの成立機序を解明し、疾患特異的免疫応答の制御法・治療法の確立へ向けた研究を実施してきた。まず疾患特異的な病原性T細胞エピトープを解明し、その抗原ペプチドを用いて自己抗原反応性T細胞クローンを樹立し、正常動物への細胞移入により病態誘導が可能であることを明らかにした。さらに、自己抗原120KDα-フォドリンは唾液腺上皮細胞のアポトーシスに伴うカスパーゼの活性化によって分断化をうけ病因抗原として機能発現する可能性を明らかにするとともに、カスパーゼに対する特異的阻害剤をin vivoへ投与することにより病態発症をブロック出来ることを確認した。疾患モデルにカテプシンSインヒビターを投与することにより自己免疫病変が著明に抑制されたことから、抗原ペプチドのプロセッシング機構を阻止することによるシェーグレン症候群の治療の有効性が示唆された。また、自己免疫疾患は閉経期以降の女性に極めて高い頻度で発症することから、発症に関与する性差の分子メカニズムを解析してきた。エストロジェンを介したアポトーシス誘導因子の存在が想定されたことから、Differential Display法を用いて解析し、RB関連蛋白RbAp48遺伝子が同定された。また、原因不明の難病ウイリス動脈輪閉塞症(モヤモヤ病)患者血清中に高頻度に分断化された膜骨格蛋白α-フォドリンが検出され、血管内皮細胞におけるアポトーシスが病態形成に関与している可能性が初めて明らかにされた。
