自己免疫性甲状腺疾患(橋本病)の疾患感受性遺伝子の同定とその機能解析

研究概要	本研究では自己免疫性甲状腺疾患であるGrave病(GD)および橋本病(HT)の疾患感受性遺伝子を同定し、自己免疫疾患発症の分子機構を解明することを目的としている。AITDの疾患感受性遺伝子同定のための第1段階として、約400個のマイクロサテライトマーカーを用いた全ゲノムスキャンを、AITDの罹患同胞対を対象として行い、これまでに合計14個の染色体の19個の領域にMLS>1(連鎖の可能性が示唆される)となる領域を検出した。その内、本年度は、8番染色体長腕の8q23-q24領域を中心に解析を進めた。罹患同胞対法では疾患遺伝子座を狭小化することはできないので、患者対照群による相関解析によりnarrow downすることが必要となる。8q23-q24領域の約21Mbについて、ドラフトシークエンスをもとにマイクロサテライトマーカーを検出し、最終的に多型性を示した330個のマイクロサテライトマーカーにより橋本病103例、対照群288例を対象として、アリルによる相関解析を行った。その結果、約30個のマーカーがP<0.05を示したが、あるcontigに存在するマーカーのアリルにおいて非常に強い相関を示した。そのアリル頻度は対照群で9.9%、HTで24.4%、P=10^<-5>、オッズ比=2.9(95%CI:1.8-4.8)であった。更にこのマーカーの存在する近傍約400kbにおいて、データベース上より、SNPを抽出し、最終的に36個のSNPsを用いて、相関解析を行った。4つのSNPs(40-2,23-3,21-6,21-2)で非常に強い相関を示し、さらにこれらのSNPsは連鎖不平衡にあることが判明した。これらの結果は、疾患感受性遺伝子がこの連鎖不平衡を示す約50kbの領域に存在することを示唆するものと考えられる。現在は、この約50kb領域に存在する遺伝子の同定とその構造解析を行っている。
