ユビキチン様タンパク質結合系の解析と生理的役割の解明

研究概要	酵母のオートファジーに必須な2つのユビキチン様蛋白質の機能解析をさらに進めた。本年度に得られた主な成果は以下の通りである。ユビキチン様蛋白質、Apg12はApg5と共有結合を形成するが、この結合体はApg16によって約300kDの複合体を形成することによって始めて機能し得ることが明らかとなった。第2のユビキチン様蛋白質、Apg8は前駆体として合成され、Apg4によって末端アルギニンが切断され、グリシンを露出する。Apg12と共通の活性化酵素Apg7により活性化され、Apg3を経て膜リン脂質ホスファチジルエタノールアミンに結合する。Apg12経路の欠損株ではApg8PEの量が著しく低下することから、この2つの経路は独立に働き得るが、相互に密接に関連している。Apg蛋白質に蛍光蛋白質を融合して細胞内における局在及び動態を観察した結果、これらの結合体が他のApg蛋白質が共存する構造体を発見し、前オートファゴソーム構造と名付けた。この構造上でオートファゴソームが形成されることも明らかとなった。Apg8の脂質化反応の機序を明らかとすることを目的として、この反応に関わる3つの因子を大腸菌内で発現し、これらの因子のみでApg8の脂質修飾が起こり得ることを証明した。さらに精製蛋白質と、ホスファチジルエタノールアミンを含むリポソームを用いた再構成に成功した。PEへの修飾にはリポソームのPE含量が重要な要因であることが明らかとなった。
