虚血性神経細胞死の分子機構解明と治療への応用

研究概要	この3年間の研究で、脳虚血に伴う遅発性神経細胞死のメカニズム解析を進めてきた。雄性Wister rat前脳虚血モデルを用い詳細な検討を行った結果、虚血性神経細胞死におけるCsAのほぼ完全な抑制効果は、(1)Bcl-2 family memberで細胞誘導蛋白であるBadを介した、カルシニューリン依存性細胞死を抑制すること、(2)ミトコンドリアに発現するprolyl cis/trans isomerase、cyclophilin D(CyPD)の抑制によるミトコンドリアの安定化、(3)CREBを介したBDNF/TrkB伝達系の増強によることを報告した。これらの機序をもとに抗虚血薬の候補を探索し、新たな薬剤FR901459を藤沢製薬との共同研究にて開発した。動物実験では著明な抗虚血作用を示し、今後臨床応用に向けた開発を行う予定である。また、今後の解析を進める上で欠かせないマウスを用いた同モデルを確立し、現在共同研究にて各種遺伝子改変マウスを用いた実験を行っている。さらには、虚血時に起こる現象として、低酸素および低グルコースに反応する転写因子としてHIF(hypoxia inducible factor)の重要性が判明し、機能解析のために酵母Two-hybrid, Three-hybrid systemを用いて、新規結合蛋白に対する遺伝子の同定に成功した。今後、HIFの各サブタイプおよび同定された遺伝子を含めたsiRNAを作製し、培養細胞を用いた標的因子のknock downおよび、マウスへの応用等を検討中である。あわせて虚血性再還流モデルにおける脳組織を用いたin situ hybridizationにて部位別、虚血の影響等におけるmRNA誘導の変化も行う。
