ラットの高精度比較ゲノムマップの構築と神経系ミュータントの原因遺伝子同定研究

研究概要	前年度に引き続き、神経系ミュータントラットの原因遺伝子をポジショナルクローニングによって同定する研究を行い、以下の成果を得た。1)dmyラットについては、前年度見出した候補遺伝子の中から、正常に転写が行われない遺伝子を見出した。2)mvラットについては、原因遺伝子変異がアトラクチン遺伝子の第1エクソンを含む6,914塩基対の欠失であることを明らかにした。3)cvdラットとhobラットについては、ネトリン受容体Unc5h3遺伝子にナンセンス変異と2塩基挿入をそれぞれ持ち、その結果、ともにネトリン-Inc5h3受容体シグナル伝達に重要な役割を持つ細胞内ドメインを欠くことを明らかにした。4)furueラットについては、原因遺伝子を第12染色体にマップした。また、病理学的に解析したところ、脊髄神経根を中心にミエリン膨化および軸索変性が認められ、末梢神経病変が示唆された。5)Shumiya Cataract Rat(SCR)の白内障の原因であるオキシドスクアレン閉環酵素遺伝子(Lss)の突然変異について詳細な解析を行ない、アミノ酸置換変異およびmRNAのスプライシング異常が相加的にSCRのLSS活性を低下させていることを明らかとした。そして、SCRにおける白内障はこの突然変異のためにレンズ水晶体でのコレステロール含量が正常の約60%以下に低下した時に発症する"閾値形質"であることが判明した。また、白内障発症を不完全ながら抑制する遺伝子が存在する範囲を第15番染色体上の25〜41cMの領域に特定した。6)Deafness Kyoto(dfk)ラットのを病理組織学的に解析したところ、内耳において血管条の変性と萎縮、有毛細胞の消失、らせん神経節ニューロンの著しい減数が認められた。また、胃では胃底腺細胞の好酸性化および線維化が観察された。
