研究概要	1.ヒト型IGG抗体の作製大腸癌細胞の癌胎児性抗原(CEA)に対する遺伝子組換えマウス型モノクローナル抗体のプラズミドに対して、定常領域をヒト型となるよう遺伝子工学により入れ換え、ヒト型抗CEAモノクローナル抗体のプラズミドを作製した。 2.IGG抗体及び抗がん剤を固定化した磁性バイオプローブの作製上記IGG抗体を結合させ、それに大腸がん細胞膜抗原が結合することを蛍光標識抗体で可視化して確かめた。 3.培養細胞を用いたガン細胞殺傷抗がん剤を磁性ビーズに固定した。それを用いて、試験管内におけるがん細胞に対する細胞殺傷性を測定したところ磁性ビーズに固定化してないフリーの抗がん剤でがん細胞を処理した場合よりも、細胞殺傷性が優位に強いことが明らかになった。 4.変動磁場によるがん細胞殺傷効果ハイパーサーミア用磁性ビーズとして、上記のPrA磁性ビーズにIGG抗体を結合したものを用いて、磁場変動を介しての発熱による細胞殺傷効果を検討した。これを、培養液中に添加したところすると比較的速やかに沈殿して細胞表面に沈着したので、磁場変動を行った所、IGG抗体結合PrA磁性ビーズとコントロールの非結合性PrA磁性ビーズとの有意差は見られなかった。我々は磁性ビーズのサイズを数nm〜数百nmの幅で制御する技術を開発することに成功したので、今後、この技術を基盤にして、ハイパーサーミア用磁性ビーズとして不可欠な諸々の条件を満たし、実用化が可能な理想的磁性ビーズの開発を目指したい。
