研究概要

ヒト体液中に存在するDNaseは遺伝的多型形質であり、有用な個人識別マーカーである。本研究では、これら酵素を中心として多数の遺伝的多型形質を検出・同定する方法を確立し、法医鑑識分野における日常検査に応用できるように迅速化・簡便化を図ることを目的とする。
1. DNase I cDNAを組込んだ哺乳類細胞発現ベクターを遺伝子導入したCOS-7細胞から発現・分泌されたDNase Iを含有する培養上清について、3段階のカラムクロマトグラフィーを用いてDNase Iを単離・精製する方法を確立した。得られた酵素標品を免疫して調製した抗DNase I特異抗体は、生体試料から調製した酵素を免疫したそれと比較して免疫学的に遜色ないものであった。従って、哺乳類細胞を用いたDNase I発現系および培養上清からの酵素の精製法は抗DNase I特異抗体作製のための有用な免疫源調製法であることが判明した。
2.今回、DNase I型判定に供する血液や体液などの生体試料中のDNase I含量の生理的変動を精査した。ソマトスタチンは多くの内分泌・外分泌器官における分泌を抑制する生理的因子である。ソマトスタチンを投与したラットでは消化管におけるDNase I遺伝子発現レベルが抑制され、それに伴い血中のDNase I活性レベルの減少が惹起された。従って、ソマトスタチンは血中DNase I活性の変動を起こす生理的因子として初めて見出されたものである。
3.今回、4種類の両生類由来のDNase Iについてその生化学・分子生物学的性状を解明した。その結果、両生類酵素はC末端にCys含量の高い約60アミノ酸残基から構成される延長領域が存在するなど、他の脊椎動物由来酵素にはみられない構造上の特徴を有すことが明らかとなった。
4.従前の研究から多型と胃がんとの関連が明らかにされたDNase Iは、ヒトおよびラットにおいて胃の主細胞に局在することが免疫組織化学的に明らかとなった。
