研究概要

ヒト体液中に分布するDNA分解酵素(DNase)は遺伝的多型形質であり、有用な個人識別マーカーである。本研究では、この酵素を中心として多数の遺伝的多型形質を検出・同定する方法を確立し、法医鑑識分野における日常検査に活用できるよう迅速化・簡便化を図ることを目的とした。
1.DNase IIは精液をはじめヒト体液に広く分布する遺伝的多型形質であるが、その多型解析に利用される特異性の高い抗体がこれまで作られていない。今回、新規な抗体スクリーニング法を利用してDNase IIに対するマウスモノクローナル抗体の作製に成功した。得られた抗体は電気泳動後の免疫ブロット法によってDNase IIを高感度に検出した。さらに、本モノクローナル抗体を使用したイムノアフィニティー法による簡便なDNase II精製法を確立した。従って、今回得られた抗DNase IIモノクローナル抗体はDNase II多型検出に応用できることが明らかとなった。
2.DNase I型判定に供される生体試料中のDNase I含量の生理的変動を精査した。その結果、主要産生臓器である膵臓に匹敵する含量のDNase Iが脳下垂体に分布すること、その含量は第二次性徴に対応して増加するような年齢依存的変動を示すこと、さらにその活性変動は遺伝子発現を介して調節されていることを見出した。従って、DNase Iには内分泌作用と何らかの相関性が存在することが示唆された。
3.尿試料から簡便かつ高収率なDNA抽出法を開発した。尿試料は本法によりDNA多型検査に供される有用な生体試料となり、ドーピングや違法薬物事犯に係わる個人識別に活用できることが明らかとなった。
4.脊椎動物由来DNase Iの分子進化学的解析から、冷血動物から温血動物への進化に伴いDNase Iの熱安定性が獲得されることを明らかにした。
