研究概要

口腔外科領域の癌におけるゲノム変化を体系的・網羅的に検索し異常の検出された領域から癌関連遺伝子を単離・同定すると共に,新規に作製するゲノムDNAマイクロアレイを用いてこれらの異常と臨床病理学的データとの関連を体系的に解析することで,多項目遺伝情報に基づく口腔領域の「癌の個性診断法」を確立することを目的として研究を推進し,以下の成果を得た。
1.口腔,咽頭,食道領域の癌におけるゲノム異常の探索と検出された異常領域からの癌関連遺伝子異常を同定・単離するため,細胞株あるいは臨床検体よりDNA等の解析材料を抽出するとともに,臨床・病理学的データの収集を開始している。
2.これらの材料を対象にCGH法・FISH法にて解析することにより複数の新しい遺伝子増幅領域が検出された。これらの領域から,既知ならびに新規の増幅の標的となる癌関連遺伝子(例えば食道扁平上皮癌における1q32増幅領域内のATF3やCENPF,あるいは9p23増幅領域内のGASClやNF1Bなど)が単離でき,さらに他の領域でも解析を進めている。一部については遺伝子の機能解析も行っている。
3.網羅的かつ高精度にゲノムDNAコピー数の変化を検出するCGH法を基盤にした高密度・高感度のゲノムDNAマイクロアレイの開発に着手した。UCSF癌研究所Gray博士との共同研究により,既製のマイクロアレイを用いた方法より優れた,1コピーの変化をも検出できるシステムが実現可能であることを確認した。ゲノムワイドな遺伝子異常のスクリーニングに実用可能な高密度ゲノムDNAマイクロアレイの作製を進めている。
