研究概要

老化促進モデルマウス(SAM)のSAMP8系は学習記憶障害を発症するがSAMP8の学習記憶障害遺伝子は未だ特定されていない。そこでSAMP8,の学習記憶障害遺伝子の遺伝様式とその存在位置を特定する目的で、正常マウスであるJF1と交配させ、遺伝分析を行った.
F2世代における記憶障害発症率は約74%であり、これは記憶障害責任遺伝子が単一の優性遺伝であることを示した。また、この学習記憶障害発症には比較的少数の遺伝子が関与しており、単一の主遺伝子の存在が考えられた。さらに、記憶障害責任遺伝子のQTL(量的形質遺伝子座)連鎖解析を行ない、1番染色体、12番染色体、13番染色体上に記憶障害責任遺伝子の存在する候補領域を同定した。現在はこの各候補領域を持つコンジェニックマウスの作製を行っている。
4血管結紮一過性脳虚血モデルラットを用い,脳虚血時に海馬で発現が変化する遺伝子の単離・同定を検討する目的で,Differential Display法を用いて検討した結果,脳虚血後発現が減少する遺伝子としてphosphatidylinositol 4-kinase(PI4-K)を単離した.免疫組織学的検討において,PI4-Kの海馬における発現は,脳虚血後に神経細胞死が認められるCA1領域で特異的に減少した.さらに,PI4-Kの過剰発現はHEK293細胞における細胞死を抑制した.これらの結果から,脳虚血時の神経細胞死に対してPI4-Kは保護的役割を演じていることが示唆された.
軽度の脳虚血を負荷すると、その後の脳虚血に耐性を獲得するが、この現象は虚血耐性現象と呼ばれている.虚血を加える前に,preconditioning処置を加えておくと,転写因子CREBの活性化が亢進していることから,大脳皮質虚血耐性現象はCREBを介したシグナルトランスダクションの増強によって生じる可能性が示唆された.
