内分泌撹乱物貭の電気化学的スクリーニング法の開発に関する研究

研究概要	近年、外因性内分泌撹乱物質(EDCs)と呼ばれる化学物質群が、生殖や免疫といった生命の根源を脅かしている事が明らかとなった。これらのEDCsの内分泌系への作用機構は様々であるが、レセプターとの結合がトリガーとなることが多い。従って、EDCsのレセプターへの結合能力を評価することは、その化学物質が内分泌撹乱を誘引する可能性があるかどうかを評価するために重要である。そこで本研究では、EDCsの電気化学的プレスクリーニング法の開発を目的とし、タンパク-リガンド相互作用の代表的な系であるアビジン-ピオチン系において、その相互作用の電気化学的検出に関する基礎的な検討を行った後、その原理を性ホルモンレセプターであるエストラゲンレセプター(ER)とエストラジオール系に適用した。この目的のために、エストラジオールを電極活性物質であるドウノマイシンでラベル化したリガンドを調整した。このラベル化したエストラジオールの電極応答はERと結合することにより大きく減少した。この電流値の減少はラベル化リガンドの17β-エストラジオール部位とERとの特異的な相互作用に起因するものである事が、エストラゲン抗体、グロブリンタンパクなどを用いた検討から明らかになった。一定量のレセプターに対してLEとEDCを競争反応させたところ、全ての試験化合物からERとの結合に起因する電極応答の増加が観測され、これによって様々の化学物質の環境ホルモン用作用に関するスクリーニングの可能性を示すことができた。さらにこの方法は、電極表面に固定化されたエストラジオールとエストラジオール抗体を用いるマーカーイオンの膜透過性の制御に基づくアッセイ法へと発展した。さらに環境ホルモン物質については、スクリーニング法とは別に、その電気化学的除去法及び分解法についても検討し、カーボンファイバー電極及び酸化スズ電極を用いる新しい電気化学的手法を提案することができた
