新規脳分子であるニューロステロイドによる本能行動のモティベーション制御

研究概要

動物を本能行動に駆り立てるのがモティベーションであるが、その実体は不明である。本研究はモティベーションの制御機構を明らかにすることを目的として、我々が最近見いだした新規脳分子であるニューロステロイドに着目して研究を実施した。
本能行動の中枢でありモテイベーションを制御する脳領域(間脳の視索前野・視床下部)では、チトクロームP450scc、3β-HSD、5β-還元酵素、チトクロームP45017α,lyase、17β-HSDなどのさまざまなニューロステロイド合成酵素が発現しており、コレステロールからプレグネノロン、プレグネノロン硫酸エステル、プロゲステロン、プロゲステロン代謝ステロイド、アンドロステンジオン、テストステロンなどの多くのニューロステロイドが合成されることが明らかになった。プロゲステロンとテストステロンは生殖腺が合成する性ステロイドとして知られているが、モティベーションを制御する間脳の視索前野・視床下部でも性ステロイドが合成される事実は大変重要な発見である。さらに、この脳領域では新規のニューロステロイドが多く合成されていることが明らかになった。この新規ニューロステロイドは脳で特異的に合成される7α-プルグネノロンであった。
次に、これらのニューロステロイドの生理的変動を解析したところ、ニューロステロイドは明確な季節変動と日内変動を示すことが見いだされた。ニューステロイドの変動と繁殖活動周期に類似性があり、ニューロステロイドは視索前野のニューロン活動を高める作用があることが明らかになった。この作用を詳しく解析したところ、ニューロステロイドは膜受容体を介したnon-genomic作用によりニューロン活動を制御することが示された。新しく見いだされた新規のニューロステロイドにはその作用が強いことが示された。従って、ニューロステロイドは視索前野のニューロン活動に変化を導くことで、繁殖行動のモティベーションを制御すると考えられる。
