研究概要	我々は、シリケンイモリ腹腺のcDNAライブラリーよりアカハライモリの雌誘引フェロモン、ソデフリンに相当する分子をコードする遺伝子のクローニングを行い、シリケンイモリはアカハライモリのソデフリンとアミノ酸2残基を異にするフェロモンを有することを推定した。本年その物質の単離を試み、成功した。同物質はシリケンイモリ雌に対して誘引活性を示すが、アカハライモリに対しては誘引活性を持たぬことも証明され、生殖隔離現象にフェロモンも関与していることが示唆された。同物質はシリフリンと命名され、合成シリフリンを用いて抗体の作製と、ラジオイムノアッセイ系が確立された。それを用いてシリフリン含量の測定を行い、プロラクチンと雄性ホルモンの投与により腹腺中の含量が増加すること、シリフリン前躯体cDNAをプローブとしてノーザン解析とin situハイブリダイゼーションを行い、上記ホルモンにより腹腺中のシリフリン前躯体mRNAのレベルが顕著に上昇することを示した。このことからシリフリン合成にプロラクチンと雄性ホルモンの関与が明らかとなった。一方ソデフリンを嗅上皮に作用させて得られた嗅電図の解析から、鼻孔側室をとりまく上皮の一部が特にソデフリンに対して高い反応を示すことが明らかにされた。更にこの部分の細胞を酵素処理により解離し、ソデフリンを作用させ細胞内カルシウムの増加を指標としてソデフリンに反応する細胞が存在するか否かを調べた結果、一部の細胞にのみソデフリンにより細胞内カルシウムの増加が起こることが確かめられ、ソデフリン受容体を有する細胞の存在が示唆された。求愛行動の発現に関してはアルギニンヴァソトシンが発現頻度を高めるが、腹腔内投与によるより脳内投与の方がより微量で効くことを確かめ、同ホルモンが中枢に作用している可能性も示された。
