ペプチド開口放出の調節とペプチドによる神経修飾作用の分子生理学的解析

研究概要	本年度の研究では、脊椎動物においてGnRHニューロンの解析に最も適した実験系の一つである淡水産熱帯魚ドワーフグーラミーの脳を用い,まず脳・下垂体スライス標本から培養液中へのGnRH放出をラジオイムノアッセイ(RIA)で測定し、視索前野と終神経という異なるGnRH系の分泌活動の違いやそれらの機構を明らかにした。また,視索前野GnRH系においてGnRH放出の顕著な雌雄差を見出し,それがsbGnRHという分子種のGnRHペプチドを産生するニューロンの雌雄差によることを発見した。次に,微小炭素繊維電極を作製してGnRHを電気化学的に測定する方法を開発し、この方法を用いて、脳・下垂体スライスにおける視索前野GnRHニューロン軸索終末領域から、ホルモン分泌活動のリアルタイム記録を行った。また、GnRH放出に関与すると考えられるイオンチャネルなどの検索を行った。次に,GnRH放出に重要であるGnRHニューロンのペースメーカー活動の生成と,神経伝達・修飾物質によるその修飾のメカニズムについて電気生理学的に解析した。その結果,GnRHペプチド自体による終神経GnRHニューロンのペースメーカーの修飾にはNおよびR型のカルシウムチャネルが関与していることが強く示唆された。また,終神経GnRHニューロンへの興奮性シナプス入力としてグルタミン酸が候補に上がったので,このグルタミン酸受容体の同定を試みた。各種のアゴニスト・アンタゴニストを用いた実験の結果、既知のグルタミン酸受容体に加え、これまでにない新しい薬理学的性質を持ったグルタミン酸受容体があることを発見した。
