細胞死による植物生存戦略の分子遺伝学的解析

研究概要	プログラム細胞死は多細胞生物が有する基本的な生命現象の一つと考えられる。動物ではDNAの損傷、薬剤、成長ホルモン等の細胞死誘導因子によって、クロマチンの凝縮、細胞核の変形、DNAの断片化等の形態変化が誘引される。これらは厳密に制御されたシグナル伝達系を介していることから、障害等による受動的な死とは区別される。植物においては、維管束の管状要素の分化、花の形成過程に見られる雄性あるいは雌性器官の退化に伴う細胞死、花粉形成時に見られるタペート細胞の死、根冠細胞の離脱、病原菌によって引き起こされる過敏感細胞死、花弁や葉の老化等などが、プログラムされた細胞死であることが明らかとなってきた。これらは、発生分化、環境適応の過程で重要であることから、原因となる遺伝子の単離が望まれる。特に、植物のストレス応答をプログラム細胞死の面から解析する目的で、関連遺伝子の単離及び、ストレス耐性獲得機構との関係について解析が必要である。本研究では、植物のストレス応答をプログラム細胞死の面から解析する目的で、関連遺伝子の単離およびストレス耐性獲得機構との関係について解析した。イネの種子根において破生通気腔が形成される過程で発現量が変化する遺伝子の単離を行い、多数の機能遺伝子を同定した。そこで単離された遺伝子の特性を知る為、形質転換植物を作成してストレス抵抗性の変化を検定した。特に、NADPHリダクターゼを高発現する形質転換イネを解析した。その結果、イモチ病、紫外線、H_2O_2等への耐性機構を示した。すなわち、NAP代謝の活性化が主たる原因である事を証明した。
