研究概要	本研究では、イネに含まれる多種類の蛋白質を同定し、それらの機能と機能ネットワークの解明を目指す。本年度は、種子蛋白質を二次元電気泳動で分離し、発芽過程における動態を調べた。その結果、発芽過程で発現量の変動がみられる蛋白質、すなわち、発芽成長に役割を果たしていると推定される蛋白質を特定することができた。一方、翻訳後修飾と機能との関係を明らかにし、それをデータベース化する目的で、昨年度までにイネの巨大蛋白質複合体プロテアソームの翻訳後修飾を網羅的に解析し、プロテアソームサブユニットのN-アセチル化、N-ミリストイル化、リン酸化などの翻訳後修飾を検出した。そして、N-アセチル化やリン酸化とプロテアソームの蛋白質分解酵素活性に関連があることを明確にした。本年度は、プロテアソームのリン酸化が19S調節因子のATP分解酵素活性に大きな影響を及ぼすことを明らかにすることができた。また、この研究においてイネのプロテアソーム19S調節因子の6つのサブユニットをコードする遺伝子にきわめてよく似た配列をもつ2つの異なった遺伝子(重複遺伝子)が存在することを示したが、本年度、19S調節因子には、実際にこれらの2つの異なった遺伝子から翻訳された2種類の蛋白質が含まれていることを質量分析によって明らかにすることができた。これら2つの重複遺伝子産物は、組織間で発現比率が異なっていることから、組織間の機能的な違いと関連している可能性があると考えられた。なお、本年度は、蛋白質間相互作用を効率的に解析する目的で、新規なダイヤモンド様炭素被膜基板に電気泳動で分離された蛋白質をエレクトロブロッティングによって転写し、プロテインチップを作製する手法を開発した。さらに、基板へ転写された蛋白質に相互作用する蛋白質を結合させ、結合した蛋白質を質量分析装置で同定する方法を開発することができた。
