研究概要

本研究では、イネに含まれる多種類の蛋白質を同定し、それらの機能と機能ネットワークの解明を目指す。昨年度までに、種子蛋白質を二次元電気泳動で分離し、発芽過程における動態を調べ、発芽過程で発現量の変動がみられる蛋白質、すなわち、発芽成長に役割を果たしていると推定される蛋白質を特定することができた。本年度は、これらのデータを収納したデータベースを作成した。また、これまでにイネの巨大蛋白質複合体プロテアソームの翻訳後修飾を網羅的に解析し、プロテアソームサブユニットのN-アセチル化、N-ミリストイル化、リン酸化などの翻訳後修飾を検出した。そして、N-アセチル化やリン酸化とプロテアソームの蛋白質分解酵素活性に関連があること、また、プロテアソームのリン酸化が20Sプロテアソームのプロテアーゼ活性だけでなく、19S調節因子のATP分解酵素活性に大きな影響を及ぼすことを示した。本年度は得られた翻訳後修飾と機能に関する情報を入力したデータベースを作成した。一方、イネ種子胚芽の膜結合型蛋白質を網羅的に解析するため、膜画分の精製を試みた。しかし、依然精製画分には他のオルガネラが混在しており、純度の高い画分を得ることができなかった。さらに、新規なダイヤモンド様炭素被膜基板に電気泳動で分離された蛋白質をエレクトロブロッティングによって転写してプロテインチップを作製し、チップ上でイネタンパク質と相互作用するペプチド(この実験に用いたペプチドは、種子胚芽タンパク質のリシルエンドペプチダーゼ消化によって得た)の検出を試みた。その結果、ダイヤモンド様炭素被膜基板を用いて蛋白質-ペプチド相互作用を解析できることを示すことができた。
