イネ葉の老化に伴う機能衰退の分子機構-Rubisco合成能低下の要因解析-

研究概要	Rubiscoについて、イネ葉の一生を通してタンパク量の変動と単位時間当たりの生成量・分解量を調べ、それらと大、小サブユニットのmRNA量の変化、rbcS、rbcL遺伝子の量的変動、rRNA量の変動等を調べ、老化過程におけるRubisco生成の変動要因を解析した。 Rubiscoの生成は、葉の一生を通して大小サブユニットのmRNA量の変動に少し遅れながらおよそパラレルに変動した。rbcSおよびrbcLのmRANsは共に葉の展開前半に最大量となり、葉の展開終了時にはそれぞれ、最大時の三分の一、五分の一以下となった。その後はいずれも、十分の一以下にまで減少した。Rubiscoの生成は葉の展開中頃がもっとも盛んで葉の展開終了直後には最高時の四分の一以下となり、その後はさらに十分の一以下にまで減少した。一方、rbcL DNA量は葉の展開後半に最高値に達したのち除々に減少していったが、rbcL mRNAほど急激には減少せず、mRNAレベルと直接支配していなかった。Rubiscoの分解は展開終了頃には既に始まり老化初期にピークがあった。老化過程の分解量は生成量をずっと上回っていた。これらの結果より、葉の一生におけるRubisco生成の7-8割は、葉の展開終了時までに終えており、その生成量は葉の一生を通して大小サブユニットのmRNA量とおよそパラレルな関係にあることが分かった。葉の展開終了後は大小サブユニットのmRNA量、なかでもrbcS mRNA量のレベルは特に低く、展開終了後の葉ではRubisco生成のポテンシャルが大きく低下しており、葉のRubisco量は分解速度によって主に支配されていることが明らかとなった。
