枯草菌における分泌蛋白質と膜蛋白質の謝別と局在化のネットワーク

研究概要	枯草菌および類縁菌はデンプン糖化用α-アミラーゼや洗剤用プロテアーゼ、セルラーゼ生産菌として産業的に利用されてきた。これら有用酵素のより高い生産性や異種生物に由来する有用蛋白質の分泌生産への利用を目的として、分泌蛋白質と膜蛋白質との識別と局在化の分子機構およびそのネットワークを解析した。 (1)枯草菌における高いタンパク質分泌は前駆体タンパク質の認識過程で作するSRP-SRP受容体系とタンパク質膜透過で中心的な働きをするSec蛋白質群との共同作用で効率良く行われている。SRP-SRP受容体系とSec蛋白質群のジャンクションをin vitro系を開発して研究した。SRPの構成成分であるFfhとSecAとの間で前駆体タンパク質が受け渡されることを明らかにした。さらにヒトSRPの受容体の相同因子であるFtsYはSRPとSecAの解離に関係している可能性があることを示した。 (2)コンピュータソフトウェアSOSUIおよびP-SORTを利用して、枯草菌の全遺伝子産物4,100種類を解析し、その1/4にあたる約1000種類が細胞膜タンパク質であると推定し、種々の可溶化条件で細胞膜タンパク質を抽出し、一次元および二次元ゲル電気泳動法で解析した。さらに分離したタンパク質を解析するためにPVDFメンブレンを利用した"On membrane digestion"法を開発し、MULDI-TOF MSによって約650種類の蛋白質を同定した。
