樹木の重力刺激対応とあて材形成機構に関する基礎的研究

研究概要	針葉樹のメタセコイア(Metaseqouia glyptostroboides)苗木を用いて、圧縮あて材形成におけるオーキシンとエチレンの関与について明らかにした。実験には8年生の実生苗木を用いた。6月20日、苗木を45°の角度に傾斜させた。2週間後、傾斜苗木の一部を収穫したのち、残余を反対側に逆転させ、45°の傾斜角で固定した。その後2週間育成し、7月18日にすべて収穫した。幹の一部は組織構造の解析に用い、その他は-80℃に保存し、内生オーキシン(インドール酢酸=IAA)の定量とエチレン放出量の測定に用いた。オーキシンの分析には、定法による抽出操作ののち、日本女子大学のガスクロマトグラフ質量分析計(GC-MS)を用いて定量分析した。組織構造の観察結果から、傾斜をまったく逆に変化させることで、圧縮あて材形成の場が顕著に変化し、常に傾斜した幹の下側で急激な細胞分裂が生じることを確認した。さらに内生IAA量とエチレン放出量についても、幹の傾斜角の逆転とともに変化が認められた。幹を逆転する以前には、下側に位置する幹の形成層付近における内生オーキシン量とエチレン放出量は上側に比べて高い値を示した。一方、幹の逆転処理後には、新たに下側となった部位での急激なIAAとエチレンの増加を確認した。これらの結果から、圧縮あて材形成には、傾斜した幹の下側におけるオーキシンとエチレンの増加が常に不可欠であることが予想された。またサクラ属の樹種などを用いて、微重力下における成長反応、重力感応におけるデンプン粒の役割、しだれ現象とGA, IAA,およびABAの分布などについても明らかにした。さらに樹木に対するジベレリン処理が広葉樹引張りあて材の誘導や微小管性質の変化におよぼす影響などについても新しい知見を得た。最後に、これまでの知見をまとめて、報告書として取りまとめた。
