魚類の始原生殖細胞の特性解析とその生殖工学的研究

研究概要

1)キンギョの受精直後からPGCsが生殖隆起に至るまでの正常発生過程におけるvasa遺伝子産物の分布を明らかにし、ゼブラフィッシュと異なるいくつかの知見を得た。2)機能的なPGCsの形成には、中胚葉誘導を必要としないことを実験発生学的に明らかにした。3)キンギョの胞胚から嚢胚にかけてのPGCsの移動は、形態形成運動に依存することを実験発生学的手法を用いて明らかにした。4)胚盤の移植により胞胚期にPGCsの分布を動物極側に変更した胚で、その後のPGCsの分布を解析することで、PGCsは胞胚期での初期分布により移動経路が異なることを示した。5)胞胚期のPGCsを含む領域の移植により生殖系列キメラを確実に生み出す方法を確立した。この生殖系列キメラ個体から宿主とドナー両方の子孫が得られた。6)不妊となるキンギョ雌×コイ雄の雑種胚を宿主にすることで、ドナー由来の精子のみが作られることを明らかにした。7)vasa-GFP遺伝子導入によりPGCsを可視化したニジマスの系統化に成功した。8)このニジマス系統からフローサイトメーターを用いて大量にPGCsを精製する技法を開発した。9)ニジマス孵化稚魚の生殖隆起と他の組織間でのサブトラクション法により、PGCsを含む生殖系列に特異的に発現する遺伝子群を単離した。10)単離したニジマスPGCsをニジマス或いはヤマメ孵化稚魚に移植することで、宿主生殖腺内に移植PGCsを取り込ませ、増殖、分化させる技術を開発した。
