魚類の性分化におけるアポトーシス(自殺細胞死)の役割

研究概要	本年度では、まず、ゼブラフィッシュおよびヒラメ胚を用いて,ストレス処理およびアポトーシス誘導剤C2セラミドの投与によるアポトーシス誘導を観察した。胚のTUNEL染色およびカスパーゼ活性によってアポトーシスの高感度検出が可能であった。また、魚類細胞の主要なアポトーシス実行分子カスパーゼをクローン化した。この分子を過剰発現させた胚では顕著なアポトーシスが誘導されたことから、魚類胚の実験系はin vivoでのアポトーシスのモデル系として利用可能である。一方、ペヘレイを用いて高温起因のアポトーシスが原因と思われる生殖細胞消失過程と性分化の関係を調べた結果、雌のみ発生する低水温(17℃)では性分化期の後までに異常を示す生殖細胞が認められなかったのに対して、高水温(25と29℃)で異常を示す細胞が温度に比例して増え、生殖細胞が完全に消失した個体まで出現した。なお、雄のみが発生する29℃では孵化後1週間から生殖細胞の消失が著しいことから、生殖細胞の消失と性分化との関係が示唆された。また、アポトーシスを特異的に検出するTUNEL染色法で観察した結果、ペヘレイの性分化時期前の生殖腺でアポトーシスは温度に比例して起きていることが分かった。また、生理状態によって生殖腺でアポトーシス誘導作用を示すGnRH(生殖腺刺激ホルモン放出ホルモン)の分泌を免疫組織学的手法で調べたが、性分化期において既に分泌が確認され、性分化との関連が示唆された。一方、性決定の遺伝的マーカーの探索については、性決定において遺伝的な支配が強いパタゴニアペヘレイの性決定遺伝子を見つけるためにAmplified Fragment Length Polymorphism(AFLPマーカー)による連鎖解析を行なった。64セットのプライマーを用いた解析の結果、雄に特異的なDNA断片を増幅するマーカーACG/CAA217の開発に成功した。
