カゼインmRNAポリAの調節機構と乳腺の老化に関する研究

研究概要	マウスの乳腺では、分娩の直前からミルクの合成が如まり、合成量は徐々に増加し12〜13日に最大になる。以後急速に減少し18日頃に停止する。この間で細胞分裂は起こらないため、泌乳量は乳腺細胞の機能の活性を反映する。この実験系を用いてカゼインmRNAの安定性と老化に関する研究を行った。実験には妊娠13日(妊娠中期)から泌乳17日(泌乳末期)までの様々な時期のマウスを用いた。RNAを抽出し、ノーザンブロットによりカゼインmRNAを可視化した。RNAはオリゴdTの存在・非存在下で反応させ、RNase HでポリAを消化し、その差からポリAの長さを決定した。また、6時間乳腺をmRNA合成阻害剤を加えて培養し、mRNAの消失速度から半減期を測定した。βとγカゼインmRNAについて調べた。妊娠中ではポリAの長さは約50塩基であったが、カゼインの合成が始まると約100塩基となり、泌乳中期までほぽこの長さが保たれていたが、泌乳15日あるいは18日では約60塩基と短くなった。ポリAの長さは転写の段階で決まり、ポリAポリメラーゼ活性と平行して変化した。一方、カゼインmRNAの半減期においても同様な変化が見られた。βカゼインmRNAの半減期は、妊娠中約10時間であったが、妊娠18日では17時間、泌乳1日では19時間と安定となった。しかし泌乳18日では約5時間と不安定になった。 mRNAの安定性はポリAの長さと関係することを明らかにした。ポリAの長さは核の中で起こる転写の段階で決まり、乳腺の活性化と老化との関係を明らかにした。
