生殖細胞アポトーシス誘導プログラムの多様性と一様性

研究概要	哺乳類精子形成過程では、生殖細胞の高頻度なアポトーシスが知られているがその分子制御機構には不明な点が多い。本研究では、マウス精巣に於いて既に精子形成過程を阻害することが知られている種々の処理を行った障害精巣に於いて生殖細胞死の誘導機構を解析することにより生殖細胞死制御の一様な原理を見い出し、特に正常過程での生殖細胞死の調節機構への洞察を得ることを目的とした。昨年度の虚血再灌流精巣や生殖毒性物質の投与モデルに於ける生殖細胞アポトーシス誘導へのFas/FasL系の関与結果を踏まえ、本年度は、更に種々の生殖毒性物質を広範囲の濃度で作用させ生殖細胞死への影響を検討すると共に、BcL-2やBaxの発現について詳細な検討を行った.その結果、estradiol-3-benzoate (EB)投与の場合と異なり、diethylstilbestrolの投与モデルでは、その低濃度と高濃度で二層的にTUNEL陽性細胞の増大が観察され、高濃度ではFas/FasL系の関与を認めたが、低濃度では認められなかった.一方bisphenol A投与では、高濃度でEB同様TUNEL陽性細胞の増大を引き起こしたが、逆に低濃度では細胞死頻度を低下させた。これらの実験系で、Bcl-2の発現には殆ど変化がないが、Baxの細胞内での顕著な分布変化を示す細胞があり、それらはTUNEL陽性細胞と一致していた。これらの結果は、生殖細胞アポトーシス誘導機構の多様性を再確認させると共に、Baxの分布変化がいずれの場合にも一様に見られることを示している。これらの結果に関連して現在、チトクロームC及び活性型カスパーゼ3の発現についての基礎的検討を行っている。更に、これらの知見は遅れているFas遺伝子プロモーター活性化機構を検討する上でも優れた評価実験系を提供するものと思われる。
