肝臓と肝リンパにおける樹状細胞のダイナミックス

研究概要	目的(3)クッパー細胞との相互作用:ラットの樹状細胞および、前駆細胞がin vivoおよびin vitroで、クッパー細胞にN-acetylgalactosamine (GalNAc)糖鎖を介して選択的に結合することがわかった。さらに免疫染色とRT-PCRにより本細胞がGalNAc特異的マクロファージレクチンの蛋白とmRNAを保有することが証明された。以上から、樹状細胞の肝臓へのリクルート・集積は、樹状細胞とクッパー細胞の結合によって起こることが証明された。さらに、この結合は樹状細胞のGalNAc/galactose特異的レクチンを介したものであること、このレクチンが接着分子として使われるのみならず、GalNAc糖鎖抗原を摂取するための受容体としても働くことが強く示唆された(発表論文1、2番目)。目的(4)前駆細胞の肝内リクルート:ラット前駆細胞がラッテクスを静注後12時間で肝臓に集積することが明らかになった(発表論文2番目)。目的(6)肝樹状細胞のリンパ組織における抗原提示:異系ラットに肝樹状細胞を静脈内投与すると、肝リンパ節のT細胞領域に選択的に遊走した。本細胞はDNA合成を開始したホストのT細胞(Tリンパ芽球)とクラスター(ロゼット)を形成し、このロゼットのなかでホストの免疫応答(T細胞増殖性応答)が始まった。(発表論文3、4番目および発表図書)。目的(5)肝内祖樹状細胞:幼弱な樹状細胞が、異系ラット肝移植後にホストの全身のリンパ組織に遊走し、上記のロゼットを形成することがわかった(投稿準備中)。
