網膜中心動脈の血栓性閉塞・溶解による新たな虚血再潅流モデルを薬理的神経細胞死制御

研究概要	ラット大腿静脈より静注した光感受性物質ローズベンガルと網膜中心動脈起始部への緑色レーザー照射とによって同動脈内に光化学反応を惹起し、活性酸素種を発生させ、内皮細胞障害→血小板粘着→血栓形成を惹起した。血栓による血流遮断を直視下に確認し、種々タイミングで組織型プラスミノーゲンアクチベータ(tPA)を同じく大腿静脈より静注して、血栓を溶解し血流を再開させ、一過性網膜虚血モデルを作成することができた。一過性虚血惹起後に経時的にラットを屠殺しTUNEL染色法にて網膜神経細胞層のアポトーシス細胞を染色して、一過性虚血による網膜神経細胞のアポトーシス感受性の経時変化と網膜構築中のトポグラフィーを観察し評価した。虚血時間についてもtPAの投与タイミングを調節することにより、網膜細胞死に対して至適な条件を検討した。その結果、約1時間の一過性虚血により24時間後の組織化学的観察において網膜神経細胞の遅延細胞死(=アポトーシス)を再現性良く、しかも人の病態に近い病態、即ち網膜の神経節細胞層および内核細胞層に比較的限局して視神経乳頭に比較的近い部分に神経細胞死を観察できるモデルを確立することができた。また、マウスにて前眼房にカニュレーションを行い、それを通して動脈圧以上の静水圧を掛けて網膜虚血を惹起し、それを開放して再潅流を行い、網膜神経細胞死を同様に再現性良く惹起するモデルも確立した。更にその神経細胞アポトーシスに対する低体温の効果を確認した。この網膜神経細胞死はスナネズミ両総頚動脈結紮一再潅流後の海馬CA1領域錐体細胞遅延細胞死(DND)と対比しながら行った。
