心筋の機能発現におけるCD38/サイクリックADPリボース情報伝達系の役割

研究概要	病的心肥大・心不全などの心臓疾患は米国のみならず、日本における患者数も増加傾向にある。これら本態性心臓疾患の原因は、一部の遺伝子異常によるものを除いて不明な点が多く、その基礎的メカニズムの解明と新しい治療法の開発は、現代医学における最重要課題となっている。本研究では、我々が開発した、全身でCD38/サイクリックADPリボース情報伝達系を欠損する「CD38ノックアウトマウス」を用いて、この情報伝達系が心機能・心筋収縮に果たす役割を解明する。今年度は以下の実験結果を得た。 (1)CD38ノックアウトマウスおよび対照として正常マウスの心筋をとりホモジナイズして組織中のサイクリックADPリボース含量をラジオイムノアッセイ法により測定した。心筋細胞中のサイクリックADPリボース量はCD38ノックアウトマウス心筋では10〜20%程度に大きく減少していることが見いだされた。 (2)CD38ノックアウトマウスおよび対照として正常マウスの心臓の左室重量・右室重量・心房重量を測定して比較した。生後3および9か月齢において、左室はCD38ノックアウトマウスで約20%重くなっており(左室肥大)、これはCD38/サイクリックADPリボース情報伝達系の欠損による代償性変化の一部と考えられた。 (3)CD38ノックアウトマウスの心筋を組織学的に検討するため、まず光顕レベルで一本一本の心筋繊維の太さにつき、正常およびCD38ノックアウトマウス間で比較検討した。心筋線維幅はCD38ノックアウトマウスで約20%増大しており、左室肥大の原因と考えられた。 (1)〜(3)の結果から、CD38/サイクリックADPリボース情報伝達系による細胞内Ca^<2+>プールからのCa^<2+>動員が正常の心機能維持に重要な役割を果たしていることが示唆された。このように今年度の研究計画は順調に進行し、研究目標は当初の計画通りに達成された。
