細菌感染病態における一酸化窒素の多面的病因論に関する研究

研究概要	本研究においては、細菌感染における一酸化窒素(NO)の多面的な機能発現のメカニズムを解析し、NOの関わる細菌感染病態の解明と治療の新たな展開を試みる。特に本年度は、誘導型(iNOS)欠損マウスを用いて、ネズミチフス菌感染におけるNOの感染防御作用のメカニズムについて解析した。その結果、野生型マウスのネズミチフス菌感染において、本菌の主要な標的臓器である肝組織にiNOSが発現誘導され、過剰なNO産生がもたらされることがわかった。一方、iNOS欠損マウスにおいては、ネズミチフス感染に伴ったNO産生誘導は認められず、野生型マウスに比較して、ネズミチフス菌の肝組織における増殖が促進し、重度の敗血症に移行し、死亡率が著明に亢進した。さらに興味あることに、iNOS欠損マウスのネズミチフス菌感染肝組織においては、広範囲な肝細胞アポトーシスの誘導が認められ、NOがネズミチフス菌に対して直接的な抗菌活性を発揮するだけでなく、ネズミチフス菌により感染した組織や細胞(肝細胞やマクロファージなど)に対して、その強力な抗アポトーシス作用により細胞保護効果をもたらしているものと思われた。この知見より、ネズミチフス菌感染症において、NOが多面的な感染防御作用を発揮していることが明かとなった。現在、腸管におけるネズミチフス菌の侵入場所であるパイエル板におけるNOの感染防御作用のメカニズムを、その抗アポトーシス活性に焦点をあて解析を進めている。
