異物薬物排出ポンプの構造機能と多剤耐性

研究概要	緑膿菌の抗生物質多剤耐性は、この菌が発現するMexAB-OprM異物・抗生物質排出ポンプによるところが大である。このポンプの三つのサブユニットの内、MexB蛋白は内膜を12回貫通するポリトピック型トランスポーターである。このMexBサブユニットが基質を認識しエネルギーを消費して基質を認識するものと考えられている。(i)そこで先ず行った実験は、MexBの基質認識ドメインを決定する実験であった。MexBとその類似体であるMexYは、各々にβラクタムとアミノグリコシドを認識する。そこでこれらのトランスポーターの基質認識部位を明らかとするため、MexBのペリプラズム側膜外ドメインをMexYの膜貫通領域につないだ雑種蛋白を構築した。この雑種蛋白の基質特異性はMexB型でβラクタムを効率よく排出した。このことはRND-型排出ポンプがペリプラズムに突出したドメインによって基質を認識していることを示している。更に膜貫通領域が基質認識に係っている可能性を検証するため、MexBの膜貫通領域を一本一本MexYのそれと置換した。その結果これらの雑種蛋白のすべてがMexB型の基質認識を行いβラクタムを排出したけれども、アミノグリコシドの排出は認められなかった。これらの実験結果から膜貫通領域は基質認識に係っていないと結論でき、上のペリプラズム側ドメインによる基質認識の結果を支持した。この結果は異物・抗生物質認識ポンプがこれまで考えられていた様な膜を貫通しての能動輸送と全く異なる新しい型のトランスポーターであることを示した。その他の実験としては(ii)MexA及びOprMの結晶回析、(iii)MexBの膜貫通領域4と10の相互作用、(iv)MexAB-OprMポンプとDNA-gyraseによるキノロン耐性への共役関係など多くの成果が得られた。
