ウイルス誘導細胞融合とそれに対する宿主制御機構の解析-細胞融合の生物学的意義-

研究概要	ウイルスよる細胞融合誘導はウイルス側因子と宿主細胞側因子との相互作用によって制御されている。ウイルス侵入機構、膜融合及び多核巨細胞形成に関与するウイルス側因子と宿主細胞側団子を解析する。細胞融合に関与する因子としては、ウイルス誘導細胞融合の制御因子、Fusion Regulatory Factor-1(FRP-1)が重要である事を証明した。FRP-1分子は細胞融合をpositiveにもnegativeにも制御できる多機能分子であり、ウイルスとFRP-1分子との細胞相互作用を解明することにより、ウイルスの発症病理の重要な局面が明らかになると考えられる。ウイルス誘導細胞融合と生理的細胞融合の共通する分子機構を解明することにより、生物現象における細胞融合の意義を解明した。FRP-1 transgenic miceやknockout miceを作製し、FRP-1分子の生理的重要性を明確にした。ウイルスによる細胞融合は多様な因子の関与と多段階のステップを含む複合的な機構から成り立っている、FRP-1は多様な因子と共同してFRP-1複合体を形成し、生理的膜融合にも深く関わっているので、FRP-1媒介細胞融合制御の分子機構を解明することは基礎生物学的にも新たな水準を切り開いた。FRP-1のlight chainの機能を解析するともに、FRP-1のlight chainのN末端側と高いホモロジーを有する蛋白を3つ同定した。FRP-1のlight chain及びN-light chainのウイルス誘導細胞融合への作用は残された重要課題である。
