宿主因子によるパラミクソウイルスの遺伝子発現と病原性の制御に関する研究

研究概要

15年度はセンダイウイルス(SeV)V蛋白質の遺伝子発現・病原性発現と宿主因子との関わりを中心に検討を行った。V蛋白質は培養細胞における増殖には必要ないがマウスにおける活発な増殖や病原性発現に必須で、innate immunityを回避する機能が想定されている。
(1)V蛋白質の構造・機能相関:V蛋白質C端側アミノ酸配列にはパラミクソウイルス亜科ウイルス間で高度に保存された3つの領域がある。これらの領域に変異を導入してウイルスを作成し、マウス肺におけるV蛋白質の機能を検討した。V蛋白質の機能にはZn-finger-like motifを構成するCysに加えて、それ以外の保存されたアミノ酸も重要であることが明らかとなった。(Virology,2002)
(2)SeV強毒野外株病原性発現におけるV蛋白質の役割:V蛋白質の機能は卵で何代も継代された弱毒実験室株のZ株で明かにされた。そこで野外株(浜松株)V蛋白質の機能を検討したところ、意外にもV(-)浜松株はマウスにおいて浜松株と同等の増殖性と病原性を示した。しかし、Z株と浜松株のV特異領域の入れ替え実験によって、浜松株V蛋白質もZ株においてマウスにおける増殖促進効果をもつことが確認された。また、浜松株に弱毒化に関与するリーダー変異を導入したところ、浜松株V蛋白質はマウス肺におけるウイルス増殖と病原性に必要であった。以上より、SeV強毒野外株においてもV蛋白質は機能をもつこと、しかしマウス増殖性の高い野外株ではその機能は隠されていることが明らかとなった。(Virology,2003)
(3)V蛋白質と宿主因子との相互作用:V蛋白質と相互作用する宿主因子をyeast two-hybrid assayで探索したところ、Ca^<2+>/Calmodulin-dependent protein kinase kinase beta(CaMKKb)と結合すること明らかとなった。CV-1細胞に発現させたラットCaMKKbにも結合した。さらにV蛋白質はCaMKKbシグナル伝達も阻害した。今後V蛋白質とCaMKKbとの結合の生理的意味を明らかにする必要がある。
