神経、免疫、内分泌スーパーシステム間のクロストークと免疫病発症における役割

研究概要

平成12年度は、神経-内分泌系間のクロストークを分子レベルで明らかとする第一段階として、神経系の調節に重要な因子が、Th1/Th2細胞においても発現しているか否かについて検討した。RDA法を用いたサブトラクション法により、抗原特異的なTh2細胞にのみエンケファリンの前駆体であるプレプロエンケファリン(PPE)が発現することを明らかにした。Th2細胞におけるPPEmRNAの発現はIL-4依存的に誘導された。Th2細胞の抽出液にはメチオニンエンケファリンが存在することも確認されたことより、Th2細胞は神経細胞と同様に、生理活性を有したエンケファリンを産生することが証明された。
神経成長因子(NGF)ファミリーとその受容体のOVA特異的Th1/Th2細胞における遺伝子発現についても検討した。その結果、NGFとその高親和性受容体TrkAの遺伝子発現が抗CD3抗体により刺激したTh2細胞で強く誘導されること、さらにニューロトロフィン-3(NT-3)の高親和性受容体であるTrkC遺伝子がTh2細胞選択的に発現することを明らかとした。TrkC遺伝子のTh2細胞選択的な発現について、さらに詳細な検討を行った結果、TrkC遺伝子はナイーブT細胞では発現しておらず、Th2サイトカインであるIL-4によりその発現が誘導される一方、Th1サイトカインであるIFN-γによりその発現が顕著に抑制されることを明らかとした。
以上の結果から、NT-3は神経系の構築に必須であるだけでなく、免疫系、特にTh1/Th2バランスにも影響を及ぼしうる、神経-内分泌系間のクロストークに関与する分子であることが強く示唆された。今後さらに神経、免疫、内分泌のクロストークと疾病との関連性について追求して行きたいと考えている。
