経口トレランスによる抗原特異的調節性T細胞株の樹立と自己免疫疾患の治療への応用

研究概要	まず経口トレランスのメカニズムを検討した。通常経口トレランスは、抗原量の違いによりそのメカニズムに違いがあるといわれている。すなわち、少量の抗原量では調節性T細胞によるActive suppressionが誘導され、大量の抗原量ではClonal deletionもしくはAnergyが誘導されるが、両方の要素が混在することも多い。そこで本研究では、経口トレランスが誘導された未免疫マウスの腸管・リンパ節、脾臓などからT細胞亜分画を分離し、それと免疫された正常マウスの脾細胞との共培養を行い増殖反応の抑制を調べることにより、移入実験をすることなくActive suppressionの存在を確認した。この系をDBA/1マウスに誘導したコラーゲン誘導関節炎に適用し、抗原としてはII型コラーゲンのみならず、軟骨に存在する自己抗原であるgp39についても分析し、抑制効果を確認した。次に、Th3/Tr1への分化誘導因子の解析を行った。経口投与された抗原が腸管粘膜においてT細胞に提示される際に、抗原特異的な調節性T細胞が生成され、それが炎症局所へ到達しTGF-βなどの抑制性サイトカインを放出することがトレランス誘導の機序とされる。TGF-β分泌細胞(Th3)細胞の分化因子としてIL-4、TGF-などが、またネガティブな因子としてIL-12が重要であるとの報告がなされているが定見として確立していない。そこで本研究では、OVA特異的T細胞レセプタートランスジェニックマウスからナイーブなT細胞を分離し、様々なサイトカインのもとで抗原刺激を行いその分化を解析すると同時に、抗原提示細胞の条件を変化させることにより、分化後のT細胞のTGF-β分泌能を検証した。
