細胞内シグナル伝達機構かつ検討した肝類洞壁細胞機能の解析とその臨床応用

研究概要	肝線維化に関与する星細胞はPDGF、TGF-beta、acetaldehyde、endothelinがcollagen産生亢進に作用している。当該年度、PDGF、TGF-beta、acetaldehyde、endothelinによるcollagen産生亢進におけるこれらサイトカイン刺激による星細胞内シグナル伝達と、retinolによるcollagen産生抑制におけるRXR/RARの核内receptorとcorepressor、coactivatorの解析し、TNFはApoptosisを介して線維化終止に関連することを分離星細胞を用いて解析した。さらに、B型C型肝炎ウイルスの細胞内シグナル伝達を解析し、このシグナル伝達によるcollagen産生機序を明らかにしている。肝再生に於ける肝類洞内皮細胞の増殖機構の細胞内シグナルの解析のため、VEGF、EGFに係わるVEGFreceptor、KDR/Flk-1からTyrosine kinaseからのシグナル伝達としてRas-MAPKシグナル伝達の系、p38、SAPK/JNKの系、JAK/STATの系、PI3K-Akt、JAK/STATのシグナル伝達の系を解析した。また、癌細胞肝転移に関連した内皮細胞への癌細胞の接着成立機序にはinterleukin 1の関与が知られている。これら肝類洞内皮細胞増殖機構、癌細胞の内皮細胞への接着の解析のため、これら細胞内のシグナル伝達を解析するため細胞内シグナルに係わる各種dominant negative遺伝子をtransfectionしたのちluciferase assayにてシグナルの伝達の検討を行なった。
