胆汁膵外分泌、胃機能調節におけるCCK受容体機能分担と遺伝子発現調節

研究概要

ヒトCCK-A受容体mRNAを測定するためcompetitive PCR法を確立し、膵臓のmRNAの定量をおこなった。ヒト切除膵臓では、10例中9例が測定限界以下であり、ヒト膵臓におけるCCK-A受容体の生理的機能は、殆度期待できないと判断した。
ヒト小腸におけるI^<125>CCK-8を用いたmicroautographyでは、小腸粘膜に結合がみられた。これらの結果から、ヒトにおけるCCKの膵酵素分泌刺激作用は、直接膵臓に働くのではなく、小腸(神経)を介した間接的作用であることを確認した。
ヒト胆嚢では、CCK-A受容体遺伝子発現はNorthern blotで十分に検出可能でN一端抗体は、CCK-A受容体をWestern blotで検出可能であった。
肥満と胆石を併存する14例中9例にコレシストキニン(CCK)-A受容体遺伝子のプロモーター領域に2箇所の塩基置換(-128のGがT、-81のAがG)があることがわかった。転写活性は、塩基置換によって変化しなかった。メチル化と脱メチル化の度合いによって、遺伝子発現量が変化していた。
CCK-A受容体ノックアウトマウスは、膵臓組織、酵素含有量に異常はみられず、CCK以外の刺激に対して、正常に反応した。しかし、胆嚢収縮は生じなかった。胃排出速度は、かわらなかった。胆石発生率が1年齢で、欠損マウスで高かった。
