神経変性疾患の危険因子としてのミトコンドリアの蓄積するアルデヒド関連化合物の役割

研究概要	ミトコンドリアに局在するアルデヒド脱水素酵素ALDH2は、エタノール代謝においてアセトアルデヒドから酢酸への反応を触媒する。私たちは、ALDH2^*2が晩期発症型ADの危険因子であり、さらにAPO Eのε4型遺伝子保持者ではその発症リスクが相乗的に増加することを報告している。しかし、ALDH2活性の欠損が神経系に及ぼす影響についての分子生物学的解明はまだなされていない。そこで、domonant negativeに働くマウスAldh2遺伝子の欠損型変異遺伝子を作製し、これをPC12細胞に導入することでALDH2活性欠損株を樹立した。ついで、4-ヒドロキシノネナール(HNE)にたいする生存維持効果を調べた。HNEは脂質の過酸化によって生じるアルデヒド誘導体で、ADなどの神経変性疾患で患者脳に蓄積することが知られ、細胞内で多くの蛋白を修飾することで細胞死を誘導する。親株や活性型Aldh2遺伝子を導入した株は10uM HNE添加24時間後でも約90%の細胞が生存していたのに対し、ALDH2活性欠損株では30-40%程度の細胞のみが生存しているだけであった。次いで、HNEが蓄積する要因となる酸化ストレスにたいする抵抗性を調べた。アンチマイシンAにより酸化ストレスを与えるとALDH2活性欠損株ではHNEの細胞内蓄積が促進され、親株や活性型遺伝子導入株と比べて顕著な細胞死が引き起こされた。以上の結果は、ALDH2の欠損により、酸化ストレスによって生じるHNEなどのアルデヒド類が蓄積して、神経細胞死が促進されていることを示している。この細胞内機構がADの発症と密接に関わっているものと考えられる。
