強皮症皮膚硬化機構の解析

研究概要

われわれはこれまでに、ブレオマイシン(BLM)をマウスの皮下に頻回に局所投与することにより、皮膚硬化を誘導することに成功した(J Invest Dermatol 1999 ; 112 : 456-462, J Rheumatol 1199 ; 26 : 2628-2634, Arch Dermatol Res 2000 ; 292 : 535-541)。培養線維芽細胞を用いたin vitroの実験においても、BLMが線維芽細胞からのextracellular matrixの遺伝子発現を増強し、BLMが線維芽細胞に直接的に作用して線維化を誘導すること、さらにその機序としてTGF-βやCTGFを介する可能性を報告した(Arch Dermatol Res 2000 ; 292 : 556-561)。このモデルマウスを用いたin vivoの検討の結果、BLMによる皮膚硬化の誘導はTリンパ球を欠くSCID mouseにも誘導されT細胞に依存せずに起こりうることを報告した(J Invest Dermatol 2001 ; 117 : 999-1001)。今年度の研究結果として、BLMにより誘導される皮膚硬化病変部の線維芽細胞は、α-smooth musle actin陽性の筋線維芽細胞への形質転換が確認され、抗TGF-β抗体で抑制されることより、TGF-βを介して誘導されることが示唆された(Clin Immunol 2002 ; 102 : 77-83)。
