造血幹細胞の増殖分化の細胞内制御機構に関する研究

研究概要	野生型(WT)KITの細胞内領域に存在する個々のTyr残基(22個)をPheに置換した変異KITを作製し、IL-3依存性細胞株BaF3細胞へ導入しSCFによる細胞増殖や走化性に及ぼす影響を検討した。KIT-WTを発現するBaF3細胞をSCFで刺激すると濃度依存性に細胞増殖や細胞運動が促進され細胞内Ca^<2+>の動員が生じた。個々のTyrをPheに置換したKITでは、Tyr567PheとTyr719Phe置換により著明に細胞運動の低下が認められた。種々のdominant negative変異体や阻害剤を用いた検討によりKITを介する細胞運動の制御には、Tyr567からSrc型チロシンキナーゼ、p38MAPキナーゼを経てErk1/2を介する経路とTyr719からphosphatidylinositol 3´ kinase(PI3K)を介する経路の2経路が重要な役割を担っていることが明かとなった。また、KIT-WTのTyr-Phe置換は、SCFによる細胞増殖には著明な影響を及ぼさなかった。また、造血幹細胞がら巨核球への分化機構についても解析した。AIM-1とSTK15(AIK)は共にG2/M期特異的に発現するセリン・スレオニンキナーゼであり、AIM-1は細胞質分裂に、STK15は分裂後期の染色体分離に関与すると考えられている。造血幹細胞が巨核球へと多倍体化時にはAIM-1、STK15の発現は持続性に低下していた。ヒト赤白血病細胞株K562や巨核球性白血病細胞株CMKにLac-inducible Systemを用いて正常型(WT)AIM-1、STK15を発現させるとWT-AIM-1ではTPAによる多倍体化が抑制されたがWT-STK15では抑制されなかった。またキナーゼ活性を持たないdominant-negative(K/R)型のK/R-AIM1を発現させると多倍体化が誘導されたがK/R-STK15を発現させても明らかな影響は認められなかった。以上のことより巨核球が多倍体化する際にはAIM-1の持続的な発現低下が重要であることが示唆された。
