赤血球膜病態における遺伝子発現制御と形質表出に関する細胞遺伝学的・分子電顕的研究

研究概要

(I)我が国における赤血球膜異常症のphenotypeとgenotypeの検索
(1)赤血球膜異常症計810家系1010症例について、臨床血液学的、膜生化学的、分子電顕的検索を行い、その病態としてのphenotype、および病因としての遺伝子解析を行い、そのgenotypeを同定した。
(2)本年度は特に遺伝性球状赤血球症の病因としてのankyrin遺伝子変異に注目し、本症の病因としてのankyrin遺伝子変異として13種を同定した。その内訳はnonsense変異4種、frameshift変異6種、splicing変異3種である。これらの変異は全て日本人に固有な変異であった。また、正常日本人164 allele、本症症例116 alleleについて当該遺伝子のpolymorphismを検索し、missense変異2種、silent変異15種の計17種を同定し、その成績をドイツ人白人本症症例246 alleleと比較検討したところ、このうち7種が日本人に固有と判明した。
(II)遺伝子発現調節因子としての遺伝子promoter領域のgene methylationの意義
赤血球膜蛋白関連遺伝子3種(β-spectrin:SPTB、band3:EPB3、protein4.2:ELB42)におけるepigenetic controlとしてのgene methylationを検討した。その結果、(1)methyl化を受ける5'-CpG-3' sites数は、SPTBでは最も多く、EPB3では中等度であり、ELB42では最も少なかった。この5'-CpG-3' sitesはSPTBでは完全にunmethylatedであるのに対して、EPB3では約2/3のmethyl化度、ELB42では、中等度のmethyl化を示した。(2)樹立培養細胞(UT-7/EPO)では、さらにELB42でも完全にunmethylatedであった。(3)したがって膜構造のmorphogenesis上、このP4.2の発現はgene methylationと共軛している可能性が高い。
現在、この領域について研究を鋭意遂行中である。
