Genotoxic stressを負荷した細胞移植による免疫寛容の誘導

研究概要

WSラットの膵島を1.0、3.2、10、32、50、100、320μg/mlの濃度のMitomycin C(MMC)で30分間培養し、その後20時間37℃で培養後、ストレプトゾトシン糖尿病C57BL/6マウスの腎皮膜下に移植する系において,その生着延長効果と、血糖制御能における影響を検討した。MMCの濃度10、32、50、100μg/mlの処理により、対照群の平均生着期間約12日に対し、23、18、26、27日と有意な生着延長効果がみられた。また、移植後の血糖制御能の障害は10μg/mlでは認められなかった。同様(10μg/ml)のMMC処置をおこなった膵島を、腎被膜下ではなく、臨床例で行われている門脈内に移植するモデルにおいても、有意な生着延長効果が得られた。さらに、BALB/cからC57BL/6のマウス同種移植の系においては10μg/mlの濃度のMMC処置のみにより、100日以上の長期生着例が得られた。すなわち、同種門脈内移植がおこなわれている臨床モデルにおいても、MMCによる膵島処置のみによって長期の生着延長効果を期待できることが示唆された。
異種移植移植部位での免疫反応を免疫組織学的に比較検討したところ、対照群では好中球、マクロファージを始め、T細胞、B細胞とともに、びまん性の線維芽細胞の出現を認めるのに対し、MMC処置群ではこれら炎症細胞の浸潤は著明に抑えられていた。膵島のTGF-β,TNF-α,IL-1β,IL-6の発現をRT-PCR法にて測定したところ、MMC処置群では未処置群に比しTGF-βのmRNAの有意な上昇を認め、また、免疫組織染色でTGF-βの発現が移植後1日目に強いことが明らかとなった。
