研究概要

侵襲微生物をパターン認識して生体防御を担当するinnate immunity(自然免疫)系、特にToll-like receptor(TLR)系の解明が急速に進んでいる。申請者らは各種菌体成分の自然免疫系による認識機構、特に歯周組織構成細胞による認識機構を研究し、口膣粘膜疾患との係わりを探ろうとした。本年度論文発表した主な成果は次の通りである。(1)一般に膜CD14(mCD14)を欠く上皮系細胞は遊離型CD14(sCD14)存在下ではTLRを介して各種菌体成分に応答する。しかし、口膣上皮細胞はsCD14存在下でもリポ多糖(LPS)、ペプチドグリカン(PGN)ならびにその活性中心に当る合成ムラミルジペプチド(MDP)、グラム陽性菌のリポタイコ酸(LTA)に不応答である。これは、常に多量の細菌に晒されている口膣上皮細胞にあっては合理的な仕組みと考えられる。(2)MDPはヒト単球系細胞をCD14,TLR2および4非依存的に作用してMyD88発現を増強し、LPSやLTA(いずれもTLR4依存的活性を示す)と相乗的なサイトカイン誘導作用を発揮する。(3)インターフェロンγで処理された歯肉線維芽細胞はmCD14ならびにMyD88発現が上昇して、LPS高応答性を示すようになる。(4)Micrococcus luteus細胞壁ダイクロン酸はCD14およびTLR4依存的な活性を示す。申請者らは研究対象をさらに広げて、(4)innate immunityのもう一つの重要な担い手である補体系特にレクチン経路が、マンノースホモポリマーを有するLPSによって強力に活性化され、マウス肺での血小板崩壊惹起してマウスをショック死に導くことを明らかにした。
今後、さらに多くの菌体成分と自然免疫系との係わりを解明し、新しい視点から歯周病研究に資したいと考えている。
