歯胚と歯原性腫瘍の分子病理学的研究

研究概要

器官形成の上皮間葉相互作用に関わる因子として注目されているBMPファミリーが,歯胚分化過程のいかなる時期で発現しているかをin situ hybridization法を用いて検索した。胎生11日にすでに発現が見られたのはBMP-4および7であり,胎生12日になるとBMP-2の発現も見られた。一方,歯胚基底膜の主要構成成分であるIV型コラーゲンのα1からα6までの6種類のα鎖の歯牙発生過程での局在と消長を免疫組織学的に観察した結果、歯胚上皮基底膜にα鎖の部位および時期特異的な局在と組み合わせが存在することを明らかになった。特に帽状期後期からの内エナメル上皮基底膜には、α1,α2,α4鎖を含む他の臓器にはない特異なα鎖の組み合わせが存在し、その組み合わせが歯胚特異的な形態形成、細胞分化に関与していることが示唆された。
歯原性腫瘍におけるIV型コラーゲンは,エナメル上皮腫ではα1,2,5,6鎖から構成されていたが,腺様歯原性腫瘍,エナメル上皮線維腫ではα4鎖の特異的な局在が見られた。歯原性腫瘍においても歯胚と同様に特異なα鎖の局在が腫瘍細胞の分化に関与していることが考えられた。
Cbfa1ノックアウトマウスにおける臼歯の歯胚発育は、組織学的には帽状期で停止しており,IV型コラーゲンα鎖の分布もα5鎖が特異な染色性を示すなどの所見が認められた。一方,切歯では基質蛋白の遺伝子発現の面からは象牙芽細胞およびエナメル芽細胞にまで分化しており,臼歯とは異なるシグナル伝達系が存在する可能性が示唆された。
