研究概要

我々は我々の開発した4-nitroquinoline 1-oxide(4NQO)誘発ラット舌癌モデルを用いてマツピングを試みたところ、舌癌発生を促進または抑制する複数の遺伝子を見出すことができた。そこで我々はspeed congenic法により上記の遺伝子を基準ラットゲノムに導入し、それぞれの遺伝子の生物学的作用の解明とポジショナルクローニングを行っている。この方法で、舌癌発生を促進する遺伝子はNQO1(DT-diaphorase)である可能性が高くなったと考えられ、ヒトについても本遺伝子の多形が舌癌を含む口腔癌の発生に極めて関連が高いことが示された(癌学会、2000)。
別に、上記のラット舌癌発生に関連する遺伝子について、ヘテロ接合生の消失(LOH)及び点遺伝子突然変異の有無を検索し、成果を発表した(病理学会、2000口頭発表及び論文作成=投稿中)。
さらに、ラットの主要組織適合抗原遺伝子群が座位をしめる、第20番染色体について舌癌発生に関連するQTLの有無を検索している。この染色体については、これまでプライマーの開発の遅れで、検索が充分にされていなかったものである。
今後は、DNAチップの作製にとりかかり、その解析能力の有用性について、検討を開始する。
