プロテオミクス解析システムの構築に関する研究

研究概要	ヒトゲノムのドラフトシークエンスが公表され、本格的なポストゲノム時代が到来した。遺伝子は、蛋白質に翻訳されてはじめて機能を発現するうえ、翻訳後修飾が機能に密接に関与することが少なくなく、蛋白質の構造や機能を解析するプロテオミクスによる生命の再構築に多大な関心が寄せられている。本研究では、微量な蛋白質でダイナミックな変動や、相互作用を解析するプロテオミクス手法の構築を目的とし、前年度構築した手法を用いて以下の成果を得た。大腸癌発症のプロモーター作用が指摘されているデオキシコール酸をモデル薬物として用い、これを添加した培養液を用いて細胞を培養し、ディファレンシャルディスプレーにより、刺激の有無による蛋白質パターンの比較を行った。その結果、細胞質画分において顕著な発現変動が認められ、そのうち最も再現性良く変動したスポットにつき、トリプシンによるゲル内消化後、MALDI-TOFMSを用いてマウスマッピングに付した。得られた各ペプチドの分子量をもとにペプチドマスフィンガープリンティング法を用いるデータベース検索を行ったところ、本蛋白質が酸化ストレスに関与するペルオキシレドキシンIであることが判明した。一方、低分子-蛋白質相互作用解析を目的としてアシルアデニレート並びにアシル-2-デオキシグルクロニドをアフィニティ-ラベル化剤として用い、その反応性を吟味したところ、いずれも液性のコントロールによって反応制御が可能であり、相互作用部位に選択的なラベル化が可能であることが判明した。さらに前年度構築したイムノアフィニティー抽出法を駆使して酵素消化により得たラベル化ペプチドを選択的に抽出し、ラベル化部位の構造解析も可能であることを実証した。
