HIV-TatとRNAアプタマーの複合体の立体構造決定に基づくドラッグデザイン

研究概要	HIVのTatタンパク質に非常に高い親和性で結合するRNAアプタマーに関し、これまでTatタンパク質の最も単純なアナログであるアルギニンアミド分子及びより実際的なアナログであるアルギニンリッチモチーフを含む部分ペプチドとの複合体の立体構造の解析・決定を行ってきた。今回、アルギニンリッチモチーフをさらに細分化したペプチドを5種類用意し、RNAアプタマーとの複合体の解析を行った。その結果アルギニンリッチモチーフ中のどのアルギニン残基が、RNAアプタマーと相互作用しているのかを決定する事ができた。2個(ないしは3個)のスペーサー残基を介して配置された2つのアルギニン残基が、RNAアプタマー中の2つの結合サイトと各々独立に、同時に相互作用している事がわかった。これがRNAアプタマーの非常に高い親和性をもたらしているメカニズムである。上記の解析と併行して、部分ペプチドの^<13>C^-、^<15>N^-安定同位体標識も試み、それを用いた複合体の解析も進行させた。安定同位体標識したペプチドを用いる事により、相互作用に関するより詳細な情報が得られ、2つの異なるアルギニン残基によるダブル相互作用が確認された。得られた複合体の構造は、高い親和性がもたらされているメカニズムを明らかにしたのみならず、より高い親和性を有するRNA分子への改変の指針も与えた。又複合体の構造決定により、RNAアプタマーの中でTatタンパク質との相互作用に関与しない部分も明らかとなった。これらの部位は、宿主由来の転写関連タンパク質と相互作用する事で宿主の転写を阻害してしまい、副作用を引き起こす事もわかってきた。Tatタンパク質との相互作用に関与しないこれらの部位の塩基を置換する事で、副作用の少ないRNA分子へと改変できる可能性も示された。
